Charles Don graduated in medicine from Aberdeen University in 1925, having the John Murray gold medal and scholarship as most distinguished graduate of his year. After working in Sheffield, he became a registrar at Salford Royal Hospital in 1927. In 1928 he was awarded his MD for work on toxic goitre. In 1935 he became honorary assistant physician at MRI, and was later a consultant physician. Don enjoyed a considerable reputation as a teacher, and was noted for his diagnostic skill.
